CD4+T cells regulate glucose homeostasis independent of adipose tissue dysfunction in mice

چکیده
Obesity is frequently associated with a chronic low-grade inflammation in the adipose tissue (AT) and impaired glucose homeostasis. Adipose macrophages (ATMs) have been shown to accumulate inflamed AT either by means of recruitment from blood or local proliferation. ATM proliferation activation can be stimulated TH2 cytokines, such as IL-4 IL-13, suggesting involvement CD4-positive T cells activation. Furthermore, several studies alterations metabolism. Therefore, we sought examine direct impact on activation, homeostasis using an vivo depletion model. Surprisingly, CD4 did not affect proliferation, insulin sensitivity. However, led significant improvement tolerance. In line this, found moderate disturbances pancreatic endocrine function following depletion. Hence, our data suggest that effect metabolism observed after might mediated organs other than independent inflammation.

Adipose Tissue Macrophages Function As Antigen-Presenting Cells and Regulate Adipose Tissue CD4+ T Cells in Mice
The proinflammatory activation of leukocytes in adipose tissue contributes to metabolic disease. How crosstalk between immune cells initiates and sustains adipose tissue inflammation remains an unresolved question. We have examined the hypothesis that adipose tissue macrophages (ATMs) interact with and regulate the function of T cells. Dietary obesity was shown to activate the proliferation of ...
متن کاملFAS and ACC dysfunction in visceral Adipose Tissue
Background and Aim: A great interest for determining the particular mechanisms underlying lipogenesis and adipogenesis has been raised among researchers in order to fight obesity. We aimed to investigate the gene expression of FAS and its role in regulation of lipogenesis and adipogenesis in visceral adipose tissues from obese and normal-weight subjects. materials and Methods: A total of.parti...
متن کاملBrown adipose tissue regulates glucose homeostasis and insulin sensitivity.
Brown adipose tissue (BAT) is known to function in the dissipation of chemical energy in response to cold or excess feeding, and also has the capacity to modulate energy balance. To test the hypothesis that BAT is fundamental to the regulation of glucose homeostasis, we transplanted BAT from male donor mice into the visceral cavity of age- and sex-matched recipient mice. By 8-12 weeks following...
متن کاملGlucose Homeostasis in Mice Is Transglutaminase 2 Independent
Transglutaminase type 2 (TG2) has been reported to be a candidate gene for maturity onset diabetes of the young (MODY) because three different mutations that impair TG2 transamidase activity have been found in 3 families with MODY. TG2 null (TG2(-/-)) mice have been reported to be glucose intolerant and have impaired glucose-stimulated insulin secretion (GSIS). Here we rigorously evaluated the ...
